With the exception of the early 1917 Colts, most revolvers that take 45 ACP are designed so that the ACP case head spaces on the mouth of the case, just like in the auto.   That's why when you drop an ACP into a 45 Colt, it just disappears, and may slide out the front of the cylinder.   That's why the Ruger convertibles come with two cylinders, one for rimmed, and one for rimless.   In later Colt 1917s and the Smiths, you don't have to use the half moon clips to shoot, but the extractor does't work.
I think that Tarus, and some others that have produced 380 and 9mm revolvers have made adaptions to the extractors in their revolvers to engage the semi-rimmed case.

.45 Colt rounds are rimmed and head space off the rim. .45 acp rounds head space off the mouth of the casing. .45 acp revolvers have their chambers throated so the mouth of the casing will hold the round in the cylinder without need of a moon clip and hence fire properly. They will not eject though using the ejector rod without those moon clips.

Now if someone has a DA/swing out cylinder .45 Colt revolver; moon clips will allow them to also shoot .45 acp rounds through said revolver. I had a smith bore out my .45 acp Taurus 455 ss so it will also take .45 Colts now. The stainless revolver below (455ss) will fire both .45 acp, must use moon clips now, & .45 Colt.

The .45 ACP Vaqueros headspace on the case mouth since it is a "rimless" case.

I have 2 of them and they are some of the coolest guns ever. Those fat rounds drop right into the loading gate and you can load them with. 45 magazines.

9mm Single Actions aren't quite as fun.

I used to own an old S&W Breaktop in .32 S&W "short" , single action and always thought they were the perfect sized little guns.  They're just too old and fragile to shoot a lot.   I fell in love with the old .22, .32 and .38 S&W pocket revolvers,  especially the Single Actions. They just had so much Old School class.

This is what attracted me to the NAA Minis , especially the Short and LR model. They're a modern equivalent of the guns regular guys would drop in their pocket as Mugger Repellent.

I had always wished NAA would make a slightly bigger. 32 Long version.

Extraction of the .32 ACP could be a problem. I have a Davis derringer in .32 ACP that will not extract FIRED cases. Works fine with unfired rounds but the extractor will override fired cases making it very hard to reload. Five rounds would only increase the problem.
If you have a removable cylinder like the minis & push the cases out it would not be a problem but there appears to be many people who want fast reloads.
